STUDIES ON THE TOXICITY OF POLYCHLORINATED BIPHENYLS(PCBs), AND THE EFFECT OF VITAMIN A AND SILYMARIN IN RATS

Abstract
This study was carried out to evaluate the effects of vitamin A and silymarin on the acute toxicity of polychlorinated biphenyls (PCBs, containing 42%chlorine) in rats (SpragueDawley strain). A total of 50 rats were divided into 5 groups; negative control, positive control (PCBs 200mg/kg/day), vitamin A pretreated (3, 000 I. U. /kg/day), vitamin A+silymarin. pretreated (1,500I.U./kg/day+15mg/kg/day), and silymarin pretreated(30mg/kg/day). Vitamin A and silymarin was given orally 4 hours before the administration of PCBs (200mg/kg/ day) for 5 days.
The results obtained were as follows.
In all drug-pretreated groups, increase of relative organ weight of kidneys and total lipid contents in liver, and hematological and biochemical changes (RBC, Ht, GPT, Cholesterol) due to acute poisoning of PCBs was inhibited within normal limits. In the vitamin A, and vitamin A+silymarin pretreated group, increase of 3-ALAD activities was significantly reduced. Especially, pathological changes in liver cells due to the acute poisoning of PCBs was protected to almost normal limits in the vitamin A+silymarin pretreated group.
